技術說明

Rho kinases (ROCKs)屬於serine/threonine kinase，參與許多細胞功能的調控；利用ROCK抑制劑的臨床試驗及動物實驗結果顯示，ROCK在許多心血管、神經退化疾病的發生以及癌症的轉移扮演重要角色。在哺乳類細胞中表現ROCKI及ROCKII兩種基因，然而目前並沒有可以直接分別測定細胞或組織內ROCKI及ROCKII活化程度的合適方法或試劑。本發明揭示一種用於偵測生物樣本中ROCKs活化之試劑組，可分別偵測ROCK I與ROCK II活化情形，其中該組試劑包括二抗體，分別用以辨識ROCKI胜肽序列的第1333號位置絲胺酸之磷酸化或ROCKII胜肽序列的第1366號位置絲胺酸之磷酸化。利用這些抗體來偵測ROCK的活化。經實驗結果證實利用這些ROCKI與ROCKII磷酸化專一性抗體可以分別直接偵測出ROCKI與ROCKII在細胞或組織內活化的程度。 ROCKs involve in the regulation of multiple cellular processes. Increasing number of clinical trials and animal experiments using ROCK inhibitors suggest that ROCKs activation plays an important role in the pathogenesis of many diseases. However, reagents capable of directly detecting ROCKI and ROCKII activation are lacking. In this study, we disclosed a set of agents for probing/detecting the activation of ROCKI and ROCKII individually encoding their amino acid sequence in biological samples, wherein the agents includes one antibody to specifically recognize ROCKI Ser1333 phosphorylation and one antibody specifically recognize ROCKII Ser1366 phosphorylation. Our experimental data indicated that the phosphorylation levels of ROCKI on Ser1333 and ROCKII on Ser1366 reflect their activities respectively. Thus, the activation status of ROCKI and ROCKII in cells or tissues can be determined individually by using these phospho-specific antibodies.
