技術說明

於明確、已知成分的細胞培養基中維持人類造血幹細胞，人類間葉幹細胞，未分化之人類胚胎幹細胞及人類誘導型多能性幹細胞之合成表面包含溶脹之聚乙烯醇表面及共軛至此聚乙烯醇表面之寡肽。此溶脹之聚乙烯醇可由包含乙酸乙烯酯及具有羧酸及胺基之單體聚合而成。此溶脹之聚乙烯醇可由包含乙酸乙烯酯表面硬度可借由掌控乙酸乙烯酯的交聯程度來控制。共軛至此表面之寡肽包含氨基酸序列 (a) RGD序列，或 (b) XaanProGlnValThrArgGlyAspValPheThrMetPro，其中n可為零至参之間任何整數且Xaa可為任一種胺基酸。於此發表之溶脹之聚乙烯醇表面可由伽馬射線照射消毒。 Synthetic surfaces capable of supporting of human hematopoietic stem cells, human mesenchymal stem cells, undifferentiated human embryonic stem cells and human induced pluripotent stem cells in a chemically defined medium include a swellable polyvinylalcohol layer and an oligopeptide conjugated to the swellable polyvinylalcohol layer. The swellable polyvinylalcohol may be formed by polymerizing monomers in a composition that includes vinylacetate and functional monomers containing carboxylic acid or amine. The elasticity of the swellable polyvinylalcohol may be controlled by crosslinking of the polyvinylalcohol. The conjugated oligopeptide may include an amino acid sequence of (a) RGD sequence or (b) XaanProGlnValThrArgGlyAspValPheThrMetPro, where n is an integer from 0 to 3 and where Xaa is any amino acid. Further, disclosed herein is a swellable polyvinylalcohol surface which can be sterilized by gamma irradiation.
