技術說明

蛋白標記(Protein marker)若能在西方點墨法(Western blot)中直接被目前市面上最廣泛使用的任何抗老鼠/兔子之二級抗體辨識且自動呈色於底片上，將能使實驗人員更有效率、方便且準確的進行實驗。我們在本研究中開發了一種含有老鼠及兔子抗體線性抗原決定位標籤，此標籤能被抗老鼠/兔子之二級抗體辨識且自動呈色於底片上。我們利用結構模擬軟體篩選三段老鼠(M1, M2, M3)及三段兔子(R1, R2, R3)IgG抗體Fc區域的線性抗原決定位(Linear epitope; LE)作為標籤的候選序列(IgG Fc LE)。利用西方點墨法發現老鼠(M2)及兔子(R2)抗體線性抗原決定位能成功被BL21大腸桿菌表現且可被抗老鼠/兔子之二級抗體直接辨識、自動呈色於底片上。我們將M2及R2結合成大小約15 kDa的含有老鼠及兔子抗體線性抗原決定位標籤(M&R tag)，並在其後依序接上不同分子量大小之蛋白，組成含老鼠及兔子抗體線性抗原決定位標籤之重組蛋白標記(M&R linear epitopes protein marker; M&R LE protein marker; 15-120 kDa)。利用西方點墨法發現M&R LE protein marker能成功被BL21大腸桿菌表現且可被抗老鼠/兔子之二級抗體直接辨識、自動呈現出規則的分子量大小於底片上。我們研發的含有老鼠及兔子抗體線性抗原決定位標籤具有以下優點:(1)方便性:可以伴隨著二級抗體染色自動呈色在底片上，功能相當實用。(2)準確性: 此標籤是dye-free，所以分子量準確。(3)可加熱:能使蛋白質標記標籤呈現出最準確的分子量。(4)廣泛應用性:能被任何抗老鼠/兔子之二級抗體辨識。(5)可作為陽性控制組。此標籤除可當作蛋白質標記(Protein marker)或重組蛋白質(Recombinant protein)的偵測標籤(tag)外，更可當做實驗的陽性控制組(Positive control)，使實驗人員能更方便且精準的判斷實驗結果，未來將有相當大的機會能取代目前傳統之蛋白質標籤，對合作企業其蛋白質分子量標記市場將開創無限的商機。 MW markers that can be self-revealed by any anti-mouse/rabbit secondary Abs will offer great efficacy and convenience for Western blot. Here, we develop a self-revealing molecular weight marker-MR marker, which contains the linear epitopes from Fc region of mouse and rabbit immunoglobulin G (IgG Fc LE). We fused M2 and R2 sequences to form a 15-kDa fusion protein (M&R tag), and then assembled with different recombinant proteins to generate 15 to 120 kDa self-revealing M&R LE protein marker. The M&R LE protein marker could be self-revealed by anti-M and R IgG secondary Abs on film. The linear epitope M&R tag possesses the following advantage: (1) Convenience (2) Accuracy (3) Heat-resistance (4) Wide applicable field (5) As a positive control. Collectively, this M&R tag not only can be applied for tags of recombinant protein and protein marker, but also can be positive control in proteomic experiment.
