技術說明

本發明則提供一種藉由偵測標的細胞中切絲蛋白表現量分析細胞老化之方法。切絲蛋白為一種分子量為約19 kD之蛋白質，其可與肌動蛋白絲 (actin filaments) 結合，並促進彼等與細胞移動、發育、極性或胞質分裂相關的動力學。哺乳動物中所表現之切絲蛋白可分為非肌肉切絲蛋白 (nonmuscle cofilin, n-cofilin)、肌肉切絲蛋白(muscle cofilin, m-cofilin)與ADF。切絲蛋白表現缺失會導致細胞停滯在G2/M階段，產生多核的細胞，以及造成不可逆的細胞死亡。然而，並無相關文獻曾經揭露，標的細胞中內生性切絲蛋白表現量與其細胞老化情況之關連。 The present invention discloses a concept that the expression level of cofilin may reflect the senescent condition of a cell or tissue. According to the findings in present invention, a method for determining the cellular senescent condition in a cell or tissue sample by evaluating the expression level of cofilin is provided. The detection of the expression level of cofilin is also used to screen an effective compound or composition for regulating the senescent condition in target cells.
