技術說明

阿茲海默氏症會造成持續性的認知能力下降和記憶力減退，是最常見的失智症。隨著人口老化增加，預估到2040年將增至8,000萬人。因此尋求有效遏止阿茲海默氏症進程之方法，乃當務之急。本研究發現3種新穎合成四氫吡喃醇化合物可降低∆K280 tauRD-DsRed 293細胞的tau聚集，並可抑制活性氧化物產生及凋亡蛋白酶3活性，及促進∆K280 tauRD-DsRed SH-SY5Y細胞的神經突生長。以上研究結果顯示所檢測的四氫吡喃醇化合物有應用於臨床治療阿茲海默氏症之潛能。 Alzheimer’s disease (AD) is the most prevalent form of dementia associated with progressive cognitive decline and memory loss. With aging of populations, the worldwide prevalence of AD will increase to more than 80 million by the year of 2040. Therefore, to identify potential treatment for AD is urgent. In this study, three novel synthetic tetrahydropyranol compounds were able to reduce the tau aggregation in ∆K280 tauRD-DsRed 293 cells. All of them also significantly reduced ROS level and caspase 3 activity in 293 ∆K280 tauRD-DsRed cells and improved neurite outgrowth in SH-SY5Y ∆K280 tauRD-DsRed cells. These data suggest that the test tetrahydropyranols could have therapeutic potential for AD.
