高通量微珠式濾膜盤分析法以是結合96孔濾膜盤與前製之分析物結合微珠進行競爭型分析。所有的反應步驟接在以鋁箔封膜封好之濾膜盤的孔洞中進行,反應後的清洗步驟則可在撕下鋁箔封膜後以清洗液加入過濾盤中,令反應後除了結合於微珠上的物質以外,其餘皆隨清洗液流出過濾盤,利用過濾盤與微珠的大小達到分離清洗的效果,若需收集通過過濾盤之物質則可以將過濾盤按照各孔洞的編號相對應組合在一般96孔黑盤上,藉由離心將要收集的物質接入一般96孔黑盤中。此高通量微珠式濾膜盤分析法無須較昂貴的磁珠與外加的磁場及可以具有與磁珠式分析法相同的優勢。 This high throughput bead based filter plate assay integrates functional beads and filter plate. Incubation steps are all performed after the 96-well filter plate bottom is sealed with adhesive foil films, while washing steps are easily done by running washing buffer through the adhesive foil film removed filter plate. Microh-beads are functional beads with the capability of accelerating the uniform distribution of the analyte solution. And running washing buffer through the filter, the functional beads can be retained inside the filter plate while unreacted complexes are washed out of the filter plate, therefore, fulfilling the purpose of separation. Washed out complexes can be either wasted or collected by a oriental combined 96-well plate. This high throughput bead based filter plate assay has the advantages of magnetic bead assays without the need of higher cost magnetic beads and additional magnetic field for separation.
