技術說明

白藜蘆醇以及紫檀芪是從葡萄以及小藍莓萃取出來的天然抗氧化物，本發明是要闡明一系列白藜蘆醇以及紫檀芪的衍生物做為抗頭頸癌以及肝癌的低毒性藥物，其中化合物4a是紫檀芪的衍生物，在體外活性測試中，我們發現4a對具cisplatin有抗藥性的頭頸癌細胞以及肝癌有抑制效果，同時4a在小鼠動物實驗經口服的方式投藥在低劑量即可抑制頭頸癌腫瘤之生長，25 mg/kg劑量可以抑制40%腫瘤生長，50 mg/kg可抑制50%腫瘤生長，100 mg/kg約可抑制80%腫瘤生長，而且受試小鼠的體重與行為都沒有異樣，表示毒性低微，我們預期4a可以成為抗頭頸癌及肝癌的候選藥物。 The present invention provides a series of novel derivatives of resveratrol and pterostilbene, among them, 4-(3,5-dimethoxystyryl)phenyl-3-hydroxy-2-(hydroxymethyl)-2-methylpropanoate (4a) exhibited significant anti-head and neck squamous carcinoma and hepatoma activity. Compound 4a demonstrated low oral toxicity during in vivo study. We therefore recommend that compound 4a and its analogs (4, 11~16) are useful as new anti-carcinoma and anti-hepatoma agents.
