技術說明

一種重組神經胺酸酶蛋白，其以人類新興流感野生種病毒pH1N1-NA (A/Texas/05 /2009)之胺基酸序列(SEQ ID NO: 1)為基礎。本發明之重組神經胺酸酶蛋白，其具有一細胞膜外部區域(ectodomain)，該外部區域具有至少99%等同SEQ ID NO: 1之一胺基酸序列，並在特定位置149、344、365及366以其他流感病毒之相對應胺基酸取代。本發明之重組神經胺酸酶蛋白具有交叉保護性免疫，並可作為廣效流感疫苗。 We investigated the mutant rN1NAs of pH1N1 near enzymatic-sites in contrast to wild-type rN1NAs (pH1N1 or H5N1) by using immunization strategy. Our results indicated that double mutants (I365T/S366N) and tetra mutants (I149V, N344Y and I365T/S366N) rN1NA-immunized group can induce the highest specific NA-inhibiting antibodies against homologous pH1N1 and heterosubtypic H5N1, H3N2, H7N9 virus strains. We therefore suggest that double mutants (I365T/S366N) and tetra mutants (I149V, N344Y and I365T/S366N) rN1NAs can be potential vaccine candidates for developing broadly heterosubtypic protective vaccines against influenza virus infection.
