檢測神經胺酸酶活性的方法

每年流感造成了全球數百萬的人受到嚴重的影響,而染性病原如流感病毒或細菌為主要造成全世界性流感的重要致病原因。在流感病毒表面具有神經胺酸酶蛋白 (Neuraminidase, NA),其功能主要是切除細胞受體上的唾液酸和流感病毒血球凝集素 (Hemagglutinin, HA) 之間的鍵結,以促進流感病毒的釋放及擴散。因此神經胺酸酶蛋白是重要的biomarker。申請專利是開發新的檢測法,即時追蹤病源的神經胺酸酶蛋白活性。新開發的方法檢測時無須耗時的檢體的前處理,且可用輕巧的電化學儀。所開發的試劑無須特殊儲藏仍有高檢測效率。 Neuraminidase has been found in many pathogenic bacteria and influenza viruses, and it is an important biomarker for infectious diseases. We developed a simple, rapid, sensitive, and label-free electrochemical assay to detect neuraminidase activity, by employing a newly synthesized latent probe that acts as a substrate for neuraminidase. Our assay was highly sensitive is comparable to those of other currently available sensitive methods such as ELISA and luminescence-based assays. Our probe is a non-protein reagent, and was shown to be robust, highly stable at room temperature and durable. Analysis of whole blood and serum samples containing neuraminidase revealed the reproducibility of our method in colored and cloudy samples. Our sensor of neuraminidase activity was shown to be superior to those used in previously reported methods and may thus be useful for monitoring biological status or progress in the recovery from diseases caused by pathogenic neuraminidase-producing bacteria.
