技術說明

一種重組桿狀病毒用以製備微小核糖核酸病毒類病毒顆粒，其中重組桿 狀病毒的幾丁質酶A(ChiA)基因與v-組織蛋白酶(v-cath)基因為功能損 壞，並利用強勢啟動子調控微小核糖核酸病毒之一病毒殼蛋白，以及利用 弱勢啟動子控調用以水解病毒殼蛋白的水解酶。本發明之重組桿狀病毒可 用於High Five 細胞或Sf-9 細胞以產生腸病毒類病毒顆粒，其所生產的類 病毒顆粒穩定性較佳，並且產量較先前技術具有顯著的提升。本發明亦提 供一種類病毒顆粒的製備方法。 A recombinant baculovirus is provided for preparing Picornaviridae virus-like particle, wherein ChiA and v-cath genes of the recombinant baculovirus are functionally disrupted and the recombinant baculovirus includes a Picornaviridae capsid protein gene under control of a strong promoter and a protease gene, which configured for encoding a protease for hydrolyzing the capsid protein, under control of a weak promoter. The recombinant baculovirus of the present invention may adopt High Five or Sf-9 cells for manufacturing enterovirus virus-like particles with improved stability and higher yields in comparison with the conventional art. A method for preparing virus like particle is also herein provided.
