技術說明

中樞神經發生感染時，組織傷害的程度與微生物所產生的毒素，微生物在細胞內的增生以及宿主本身的免疫反應息息相關。細菌性的感染即使有抗生素的治療，能幫助清除細菌，但感染後所引起的神經傷害後遺症，例如subdural empyema，hydrocephalus，cerebral infarctions，seizures等卻無法避免。若抗發炎反應的調控及免疫反應的終止發生失調或缺失時，則可能會造成中樞神經慢性發炎（例如multiple sclerosis），甚至神經退化（例如Alzheimer’s disease 和 Parkinson’s disease）。因此降低中樞神經在發炎反應中的損壞，防止中樞神經慢性發炎與神經退化是非常重要的。本研究藉由重組蛋白以及抗體產生技術發現重組細菌外膜蛋白A能在實驗老鼠中樞神經感染時提供保護，使其存活下來。我們相信這一重組細菌外膜蛋白A應能提供預防與治療感染性疾病不同的選擇，進而減少抗生素在臨床上的大量使用以避免抗藥性菌種的產生。 In this study, our data suggest that recombinant bacterial outer membrane protein A can provide protective effect on bacterial infection of mouse CNS. It is believed that such recombinant protein can provide an alternative approach in the prevention and treatment on bacterial infection, thus further reduce antibiotic remedy in clinic and avoid the appearance of drug-resistant strains.
