技術說明

本發明係提供一種用以檢測DNA甲基化及mRNA表現之小鼠基因晶片及其製備方法，其中，該晶片係具有複數個經篩選之含有小鼠基因啟動子而具CpG小島（CpG islands）序列之標記，二螢光探針，用以與各該標記進行DMH雜合反應（Differentiatial Methylation Hybridization），而藉由判讀雜合反應後之結果。本新式小鼠甲基化生物晶片，應用於肝癌模式小鼠、高濃度氧肺損傷小鼠動物模式、心臟受損模式小鼠、複製鼠胚於上遺傳變異分析、口腔癌模式小鼠及DMN誘導大鼠之肝損傷模式，均有良好的檢測篩選結果。在肝癌模式小鼠部份，利用此新式甲基化晶片，鑑定出受甲基化調控的DnaDJ1基因並進一步的在人類肝癌細胞株Hep3B中鑑定該基因之功能。 A mouse CpG island (MCGI) microarray was developmented to identify genome-wide methylation-silenced genes in the liver tumors, hyperoxia-induced lung injury, heart failure, cloned mice embryos, oral cancer, and DMN-induced liver fibrosis rats. Dual functional MCGI microarray was designed to exam the gene methylation and expression in various tissues from different physiologic statuses of rodents and mammals. Herein, we demonstrated a methylation-silenced DnaDJ1 gene that was identified from HBx-induced liver cancer mice and elucidated the gene functions in human Hep3B cell line.
